摘 要
头孢拉定为第一代头孢菌素类抗生素,广泛用于尿路及呼吸道感染,是目前临床应用最广泛的抗生素之一。然而,由于头孢拉定化学性质不稳定,严重地影响产品质量,甚至于影响到用药安全。
本文应用IR、UV-Vis、MS、HPLC、XRD等检测手段,采用实验、检测与理论分析相结合的方法,从光谱与结构性质、破坏性因素的影响两个层面,研究了该化合物不稳定性的原因以及破坏性因素的影响程度。
研究表明:温度和结晶水对头孢拉定影响较大,常见的金属离子中MnCl2和CoAC2会与头孢拉定发生化学反应。紫外光照和晶体形貌对于该化合物稳定性影响不大。此外,本项研究还在质谱检测的基础上,提出了全新的头孢拉定酸碱裂解机理。

Abstract
Cephradine is 1st generation of cephalosporins, which is used in the treatment of urinary and respiratory infection and is one of the most often used antibiotic. Cephradine is a labile compound,which influents quality of product and even the safety in utilization of drugs.
In this paper, With the combination of theoretical analysis and experiment and Applying spectrum techniques,such as IR, UV-Vis, MS, HPLC, XRD to Analyzing the instability mechanism and extent of destructive factors from spectral and structural properties and disruptive factors.
Research shows that: the temperature and the crystal water greater impact on cefradine, MnCl2 and CoAC2 have obviously complex reaction with Cephradine, UV irradiation and the crystal morphology has little effect for the stability of the compound. In addition, the study proposes a new fragmental mechanism of cephradine basing on mass spectrometry.
