HPLC测氨茶碱的血清药物浓度

摘要:目的 为测定人血清中氨茶碱浓度建立一个准确的高效液相色谱法,为临床开展氨茶碱血清药物浓度的监测提供重要保障。实验方法 利用日本岛津LC-20AT高效液相色谱仪来测定氨茶碱的血清药物浓度,用规格为150mm*4.6mm*5μm的ODS-C18柱为色谱柱,采用 0.8mL/min的流速,15℃的柱温,同时采用甲醇:水(22:78)为流动相,检测时波长为210nm,进样量为20μL。结果 获得血样处理办法和色谱条件选择方案,对所选方法的可行性进行相关性试验,所得测定人血清中氨茶碱血清药物浓度的色谱法的适用性得到证实,且在测定氨茶碱药物浓度时得到了较为良好的线性关系,据数据显示此方法检测时可以得到的线性范围在1.62μg/mL -37.8μg/mL(r=0.999)之间,实验方法的回收率均>95%,得到的日内和日间的精密度均<4%(n=5)。结论 在实验中探索得到的氨茶碱血清药物浓度测定方法具有可行性,能准确、灵敏的测出血清中氨茶碱的药物浓度,同时具有精密、准确、可多次重复的特点。

Method of Aminophylline concentration in serum by HPLC


Abstract: Objective Using RP-HPLC to establish an accurate means to measure the serum concentration of Aminophylline, to provide an important safeguard to carry out the monitoring of Aminophylline serum concentrations for the clinical. Experimental methods Use Shimadzu LC-20AT HPLC to measure the serum concentrations of Aminophylline, ODS-C18 column (150mm * 4.6mm * 5μm) column, and set the flow rate of 0.8mL/min, column temperature was 15 °C, using methanol: water (22:78) at the same time as the mobile phase, measuring wavelength 210nm, the injection volume is 20 ul. The experimental results From lots of experimental exploration we can get the Blood sample approach and Chromatographic conditions options. And the Correlation test about feasibility of the selected method Confirmed that the method is indeed suitable for the determination of Aminophylline serum drug concentration. And a relatively good linear relationship is obtained in the determination of the drug concentration of Aminophylline. According to the data show that this method of measuring can get the linear range 1.62μg/mL to 37.8μg/mL (r = 0.999) , the recovery rate of this method is more than 95%, and the intra-and inter precision is less than 4% (n = 5).Conclusions The measuring method of Aminophylline serum drug concentration in the experiment is feasible, it can accurately and sensitively measured the Aminophylline serum drug concentration, and it have the characters of good repeatability, high accuracy and so on
